目的探讨不同剂量低频低强度超声对耻垢分枝杆菌细胞壁通透性的影响。方法改变超声辐照耻垢分枝杆菌的声强和时间,通过流式细胞仪检测菌体内双醋酸酯(FDA)、碘化丙啶(PI)荧光强度的变化;电镜对比观察菌体超微结构的改变。结果随着超声辐照耻垢分枝杆菌剂量的增加:(1)菌体内FDA荧光强度逐渐减弱(P<0.01);(2)菌体内PI荧光强度逐渐增强(P<0.05);(3)电镜观察到细胞壁破裂、菌体内脂滴增多、形态改变等逐渐加重现象。结论一定剂量的低频低强度超声可增加耻垢分枝杆菌细胞壁的通透性。
Objective To investigate the effect of different doses of low frequency and low intensity ultrasound on cell wall permeability of M. smegmatis. Methods The intensity and time of ultrasonic irradiation of Mycobacterium smegmatis were changed. Fluorescence intensity of diacetate (FDA) and propidium iodide (PI) in the cells was detected by flow cytometry. Structural changes. Results With the increase of dose of Mycobacterium smegmatis under ultrasonic irradiation, the fluorescence intensity of FDA in the cells gradually decreased (P <0.01); (2) The fluorescence intensity of PI in the cells increased gradually (P <0.05); (3) Electron microscope observed cell wall rupture, lipid droplets within the cell increased, morphological changes gradually increased phenomenon. Conclusion A certain dose of low-frequency low-intensity ultrasound can increase the permeability of M. smegmatis cell wall.
