目的:研究人卵巢黄素化颗粒细胞中抗苗勒氏管激素(AMH)对干细胞因子(SCF)表达的影响。方法:收集15例年龄<35岁因男方因素行体外受精-胚胎移植(IVF-ET)患者的卵巢颗粒细胞,经原代培养,加入不同浓度基因重组人抗苗勒氏管激素(rhAMH),分别于培养的第4日与第6日采用实时定量PCR(RT-PCR)和免疫组织化学检测空白对照组与各实验组SCFmRNA及蛋白的表达。结果:RT-PCR和免疫组织化学均证实人卵巢黄素化颗粒细胞在不同浓度rhAMH干预后,SCFmRNA及蛋白的表达显著减少(P<0.05),其中15 ng/ml rhAMH抑制作用最明显;实验组第4日与第6日颗粒细胞表达SCFmRNA及蛋白无明显差异(P>0.05)。结论:AMH可有效抑制人卵巢黄素化颗粒细胞SCF mRNA及蛋白的表达。
Objective: To investigate the effect of anti-Müllerian hormone (AMH) on the expression of stem cell factor (SCF) in human luteinized granulosa cells. Methods: Fifteen ovarian granulosa cells of 15-year-old male patients underwent in vitro fertilization-embryo transfer (IVF-ET) were enrolled in this study. After primary culture, rhAMH, The expression of SCF mRNA and protein in the blank control group and each experimental group was detected by real-time quantitative PCR (RT-PCR) and immunohistochemistry on day 4 and day 6 respectively. Results: RT-PCR and immunohistochemistry demonstrated that the expression of SCFmRNA and protein in human luteinized ovarian granulosa cells was significantly decreased (P <0.05) after rhAMH intervention at various concentrations, with the most obvious inhibition being at 15 ng / ml rhAMH. In the experimental group There were no significant differences in SCF mRNA and protein expression between granulosa cells on day 4 and day 6 (P> 0.05). Conclusion: AMH can effectively inhibit the expression of SCF mRNA and protein in human luteinized granulosa cells.
