目的比较清肺化痰、益气活血中药对肺纤维化大鼠不同时间、不同吞噬细胞β型转化生长因子(TGF-β1)mRNA表达的影响。方法将77只雄性SPF级W istar大鼠随机分为7组,即假手术组、模型组、清肺化痰组、益气活血组、两法联合运用组、两法分阶段运用组及氢化可的松组,建立肺泡巨噬细胞(AM)、肺间质巨噬细胞(IM)细胞模型。用原位杂交方法检测TGF-β1mRNA表达水平,积分光密度法计算TGF-β1mRNA阳性面积。结果7 d时AM和28 d时IM的TGF-β1mRNA阳性细胞率和积分光密度显示,模型组较假手术组增加,差异具有极显著性(P<0.01);中药各治疗组较模型组降低,差异具有显著性(P<0.05)。结论清肺化痰、益气活血中药均可通过减少AM、IM分泌TGF-β1,抑制肺泡炎和肺纤维化的进程。
Objective To compare the effects of Qingfei Huatan, Yiqi Huoxue Chinese medicine on expression of β-type transforming growth factor (TGF-β1) mRNA in the different phagocytic cells of pulmonary fibrosis rats. Methods Seventy-seven male SPF Wistar rats were randomly divided into seven groups: sham operation group, model group, Qingfei Huatan group, Yiqi Huoxue group, two combined treatment groups, two groups of treatment groups and hydrogenation Cortisone group, established alveolar macrophages (AM), interstitial macrophage (IM) cell model. The level of TGF-β1 mRNA was detected by in situ hybridization and the positive area of TGF-β1 mRNA was calculated by integral optical density method. Results The positive rate of TGF-β1 mRNA and the integral optical density of IM on day 7 and day 28 in model group were significantly higher than those in sham-operated group (P <0.01). Compared with model group, , The difference was significant (P <0.05). Conclusion Qingfei Huatan and Yiqi Huoxue can inhibit the process of alveolitis and pulmonary fibrosis by decreasing the secretion of TGF-β1 by AM and IM.
