目的优选广痛消泡沫气雾剂中芍药苷的提取工艺。方法以芍药苷的含量为指标,采用L9(34)正交试验设计,对提取溶剂倍数(A)、提取次数(B)和提取时间(C)3个因素(每个因素设3个水平)进行考察,以确定最佳提取工艺。采用高效液相色谱法进行含量测定,Agilent extend色谱柱(4.6 mm×150 mm,5μm),流动相为乙腈-0.1%磷酸水溶液(12∶88),流速为0.8 mL/min,柱温30℃,检测波长为230 nm。结果各因素对芍药苷提取量的影响大小顺序为A>B>C,最佳提取工艺为A3B2C1即加8倍水体积,提取2次,每次1h。验证试验结果显示,芍药苷的提取重量为449.5025 mg,提取量最多。结论该工艺合理,且稳定可行,为以后的生产和质量控制奠定了基础。
Objective To optimize the extraction process of paeoniflorin in Guangtong defoaming aerosol. Methods Based on the content of paeoniflorin, orthogonal design of L9 (34) was used. Three factors (A, B, and C) To investigate to determine the best extraction process. High performance liquid chromatography (HPLC) was used to determine the content. The mobile phase consisted of acetonitrile-0.1% phosphoric acid solution (12:88), flow rate of 0.8 mL / min and column temperature of 30 ℃ , The detection wavelength is 230 nm. Results The order of influence of each factor on the extraction amount of paeoniflorin was A> B> C. The optimal extraction process was A3B2C1 plus 8 times the volume of water, extracted twice for 1 hour. Verification test results show that the extraction of paeoniflorin weight 449.5025 mg, the largest amount of extraction. Conclusion The process is reasonable, stable and feasible, and laid the foundation for future production and quality control.
