[目的]初步研究半抗原2,4-二硝基氟苯(2,4-DNFB)在体内外对肝细胞癌的抑制作用。[方法]用不同浓度的DNFB、氟尿嘧啶(5-Fu)(对照组)分别处理SMMC-7721肝癌细胞株72h,用MTT法测定72h细胞存活率,并计算50%和90%抑制浓度(IC50和IC90)。经H22肝癌细胞株皮下种植成功的18只昆明鼠随机分成3组:DNFB组、无水乙醇(Ethanol)组、磷酸盐缓冲液(PBS)组(对照组),并分别瘤内注射100μl的DNFB、Ethanol和PBS,每周2次,共4次。瘤内注射结束2周后剥离肿瘤称重,计算抑瘤率。[结果]DNFB和5-Fu对SMMC-7721肝癌细胞株的IC50分别为32.22μmol/L和3.62μmol/L;IC90分别为49.53μmol/L和>200μmol/L。DNFB组、无水乙醇组和PBS对照组最终的瘤重分别为0.97±0.53g、1.55±0.76g和3.79±0.75g。与PBS对照组比较,DNFB组和无水乙醇组的抑瘤率分别为74.43%(P=0.00004)和59.20%(P=0.00045),DNFB组和无水乙醇组之间的差异无统计学意义(P=0.16)。[结论]DNFB可在体内外抑制肝癌细胞的生长。
[Objective] To study the inhibitory effect of 2,4-dinitrofluorobenzene (2,4-DNFB) on hepatocellular carcinoma in vitro and in vivo. [Methods] SMMC-7721 hepatocellular carcinoma cell line 72h was treated with different concentrations of DNFB and 5-fluorouracil (control group), and the cell viability at 72h was determined by MTT method. Twenty-eight Kunming mice successfully subcutaneously transplanted with H22 hepatoma cell line were randomly divided into three groups: DNFB group, Ethanol group, PBS group (control group), and intratumorally injected with 100μl of DNFB , Ethanol and PBS twice a week for 4 times. Tumors were dissected 2 weeks after intratumoral injection to calculate the tumor inhibition rate. [Results] The IC50 of DNFB and 5-Fu on SMMC-7721 hepatocellular carcinoma cell lines were 32.22μmol / L and 3.62μmol / L, respectively; IC90 were 49.53μmol / L and> 200μmol / L, respectively. The final tumor weights of DNFB group, absolute ethanol group and PBS control group were 0.97 ± 0.53g, 1.55 ± 0.76g and 3.79 ± 0.75g, respectively. Compared with PBS control group, the tumor inhibition rates of DNFB group and absolute ethanol group were 74.43% (P = 0.00004) and 59.20% (P = 0.00045), respectively. There was no significant difference between DNFB group and absolute ethanol group (P = 0.16). [Conclusion] DNFB can inhibit the growth of hepatoma cells in vitro and in vivo.
