目的分析奥美沙坦对心肌肥厚大鼠心肌中MMP含量的影响及其与心肌重塑的关系。方法 60只Wistar大鼠随机分三组,模型组和干预组皮下注射异丙肾上腺素(剂量0.02mg/kg/d),对照组注射生理盐水;干预组用奥美沙坦10 mg/kg/d余两组用蒸馏水,qd灌胃;喂养8周,比较大鼠重量、心脏质量、左心室质量指数及心肌中MMP-3、TGFβ-1含量变化、大鼠心肌纤维化程度(HE染色法)及心功能情况(彩超)。结果对照组心室肥厚指标及心肌纤维化程度明显低于另两组,干预组上述指标较模型组低;心肌纤维化程度与心肌中MMP-3、TGFβ-1的水平呈正相关。结论应用奥美沙坦能够改善心功能。
Objective To analyze the effect of olmesartan on myocardial MMP in cardiac hypertrophy and its relationship with myocardial remodeling. Methods Sixty Wistar rats were randomly divided into three groups. Rats in the model group and the intervention group were injected isoproterenol subcutaneously at a dose of 0.02 mg / kg / d. The control group was injected with normal saline. The intervention group was given olmesartan 10 mg / kg / d The remaining two groups were dosed with distilled water and qd for 8 weeks. The rats’ weight, heart mass, left ventricular mass index and the content of MMP-3 and TGFβ-1 in the myocardium were compared. Results The ventricular hypertrophy index and myocardial fibrosis in control group were significantly lower than those in the other two groups. The above indexes in intervention group were lower than those in model group. The degree of myocardial fibrosis was positively correlated with the levels of MMP-3 and TGF-β1 in myocardium. Conclusion Olmesartan can improve heart function.
