用作者等所构建的重组腺病毒AdCMVgdnf直接感染多种细胞系和大鼠中脑原代神经元细胞,多重感染复数为20PFU/ml。采用免疫组织化学染色方法检测GDNF表达,观察所构建的GDNF重组腺病毒在各种细胞中介导GDNF基因转移和基因表达的能力。结果表明在一系列神经细胞或非神经细胞中,GDNF重组腺病毒均可介导GDNF基因的高效转移和表达。提示GDNF重组腺病毒可作为帕金森氏病基因治疗的高效基因转移载体。
The recombinant adenovirus AdCMVgdnf constructed by the author and the like directly infect a variety of cell lines and primary rat midbrain neurons with a multiplicity of infection of 20 PFU / ml. The expression of GDNF was detected by immunohistochemical staining, and the ability of the constructed GDNF recombinant adenovirus to mediate GDNF gene transfer and gene expression in various cells was observed. The results showed that GDNF recombinant adenovirus can mediate efficient GDNF gene transfer and expression in a series of neural cells or non-neuronal cells. These results suggest that GDNF recombinant adenovirus can be used as an efficient gene transfer vector for gene therapy of Parkinson’s disease.
