有丝分裂原激活的蛋白激酶作用激酶(MNK)对真核细胞翻译起始因子4E(e IF4E)209位丝氨酸的磷酸化作用与肿瘤相关mRNA的翻译起始过程密切相关,在肿瘤的发生、发展、转移及耐药性形成等过程中具有重要作用,MNK有望成为下一代肿瘤靶向治疗的新靶标。本文对MNK的结构、生物学功能进行综述,阐述MNK与肿瘤的关系,重点对小分子MNK抑制剂的研究进行系统总结,为新型具有高选择性的MNK抑制剂的开发提供一定指导。
The phosphorylation of mitogen-activated protein kinase kinase (MNK) at serine 209 of eukaryotic translation initiation factor 4E (eIF4E) is closely related to the translational initiation of tumor-related mRNAs, and plays an important role in tumorigenesis, development, Metastasis and drug resistance formation process plays an important role, MNK is expected to become a new target for the next generation of tumor-targeted therapies. This review summarizes the structure and biological functions of MNK, and elucidates the relationship between MNK and tumor. It mainly summarizes the research on small molecule MNK inhibitors and provides some guidance for the development of new highly selective MNK inhibitors.
