黄角颗粒对 MCAO大鼠神经细胞 bc1- 2、bax表达影响的实验研究

目的研究黄角颗粒对大鼠大脑中动脉栓塞(MCAO)模型脑神经细胞bc1-2、bax表达的影响。方法将MCAO大鼠分为中药大、中、小剂量组,西药组及模型组,另设正常组作对照;观察各组大鼠脑神经细胞bc1-2、bax表达的变化。结果模型组大鼠脑组织bc1-2、bax阳性细胞数显著高于正常组,bc1-2/bax比值显著低于正常组;中药大、中、小剂量组及西药组脑组织bax阳性细胞数显著低于模型组,bc1-2及bc1-2/bax比值显著高于模型组;中药大剂量组脑组织bc1-2、bax阳性细胞数与西药组相近,其bc1-2/bax比值显著高于西药组。结论黄角颗粒能下调MCAO大鼠脑组织促凋亡基因bax水平,上调抗凋亡基因bc1-2水平,提高bc1-2/bax比值;此可能是其抗急性缺血性神经细胞凋亡的机制之一。 Objective To investigate the effect of Huangjiao Granules on the expression of bcl-2 and bax in rat brain neuronal system (MCAO) model. Methods MCAO rats were divided into large, medium and low dose groups, western medicine group and model group, and normal group as control. The changes of bcl-2 and bax expression in rat brain cells were observed. Results The number of bcl-2 and bax positive cells in the brain of the model group was significantly higher than that of the normal group, and the ratio of bcl-2/bax was significantly lower than that of the normal group; the number of bax positive cells in the brain of the large, medium and low dose groups and western medicine group was significantly higher. Significantly lower than the model group, the ratio of bcl-2 and bcl-2/bax was significantly higher than that of the model group; the number of bcl-2 and bax positive cells in brain tissue of the high dose group was similar to that of the western medicine group, and the bc1-2/bax ratio was significantly higher. Western medicine group. Conclusion Huangjiao granula can down-regulate bax levels of brain pro-apoptosis gene in MCAO rats, up-regulate anti-apoptotic gene bcl-2 and increase bc1-2/bax ratio. This may be due to its anti-apoptosis effect on acute ischemic neurons. One of the mechanisms.
