目的探讨维康醇对DNA拓扑异构酶活性的影响。方法以质粒pBR322超螺旋DNA为底物,采用凝胶电泳分别检测维康醇对拓扑异构醇Ⅰ(TopoⅠ)和拓扑异构醇Ⅱ(TopoⅡ)介导的pBR322 DNA解旋反应的影响。结果维康醇对TopoⅠ介导的pBR322解旋反应无明显的抑制作用,而对TopoⅡ的活性有明显的抑制作用。结论维康醇的直接作用靶点可能是DNATopoⅡ,该药可能成为新的TopoⅡ抑制剂。
Objective To investigate the effect of vecon alcohol on DNA topoisomerase activity. METHODS: The plasmid pBR322 supercoiled DNA was used as substrate to detect the effect of vecon alcohol on TopoⅠand TopoⅡ-mediated pBR322 DNA unwinding reaction by gel electrophoresis. Results Weixikang on Topo Ⅰ-mediated pBR322 untwisted reaction did not significantly inhibit the Topo Ⅱ activity was significantly inhibited. Conclusion The direct target of action of Verotoxirane may be DNATopoⅡ, which may become a new TopoⅡinhibitor.
