目的:研究不同抗氧化酶在家兔动脉粥样硬化模型中的调控时效性,并进一步探讨氧化/抗氧化系统在动脉粥样硬化发病中的相互作用机制。方法:将24只日本大耳白兔随机分为正常组和实验组,实验组给以高脂饲料,行球囊拉伤术,同时静脉注射牛血清白蛋白;分别在建模0周、3周、6周、10周4个时间点耳缘静脉采血,分光光度计检测血清甘油三酯(TG)、总胆固醇(TC)、总抗氧能力(T-AOC)、过氧化氢酶(CAT),酶联免疫吸附法(ELISA)测定氧化低密度脂蛋白(oX-LDL)、谷胱甘肽过氧化物酶(GSH-Px)。结果:与正常组相比,实验组家兔氧化应激作用增强,SOD水平在第3、10周时降低,NO水平在第3、10周时增高,尤其至第10周时此水平大幅增加,T-AOC、CAT在10周末明显升高,MDA、ox-LDL水平在各时间点均增高(P<0.05或P<0.01),GSH-Px与正常组比较没有显著的统计学意义(P>0.05)。讨论:实验组家兔在动脉粥样硬化模型建立过程中,体内存在着明显的脂质过氧化损伤,氧化/抗氧化系统失衡,不同抗氧化酶可能在不同的时间窗代偿性表达,以对抗活性氧对机体的损伤。
OBJECTIVE: To study the timeliness of different antioxidant enzymes in the regulation of atherosclerosis in rabbits and to further explore the mechanism of the interaction between oxidative / anti-oxidative system and atherosclerosis. Methods: Twenty-four Japanese white rabbits were randomly divided into normal group and experimental group. The experimental group was given high-fat diet, balloon-assisted trauma and intravenous injection of bovine serum albumin. The blood samples were collected from the ears at 4 weeks, 6 weeks and 10 weeks respectively. The levels of triglyceride (TG), total cholesterol (TC), total antioxidant capacity (T-AOC) and catalase (CAT) were measured by spectrophotometer ), And oxidized low density lipoprotein (oX-LDL) and glutathione peroxidase (GSH-Px) were measured by enzyme linked immunosorbent assay (ELISA). Results: Compared with the normal group, the oxidative stress in the experimental group increased, the level of SOD decreased at 3 and 10 weeks, the level of NO increased at 3 and 10 weeks, especially at the 10th week (P <0.05 or P <0.01). There was no significant difference of GSH-Px and normal group between the two groups (P> 0.05) > 0.05). Discussion: In experimental group, there was significant lipid peroxidation injury in the model of atherosclerosis in model rabbits, the imbalance of oxidation / anti-oxidation system and different anti-oxidative enzymes may be compensated at different time windows Against reactive oxygen species damage to the body.
