目的观察通脉大生片对大鼠卵巢形态学改变及信号蛋白Smad4在TGF-β/smads通路中的表达变化及意义。方法建立大鼠肾虚排卵障碍不孕(ASR)模型,通过阴道涂片筛选造模成功大鼠后分组用药5周后,处死大鼠,HE染色观察形态学变化;蛋白免疫印迹法(Western-blot)检测大鼠卵巢中Smad4蛋白的表达量。结果 1各用药组从卵巢形态学上明显恢复的趋势,镜下可见成熟卵泡和黄体。2与模型组相比,通脉大生片高、中、低剂量组及阳性对照组大鼠卵巢Smad4的表达水平升高(P<0.05),以高剂量组及来曲唑组最明显。结论通脉大生片可用于治疗肾虚排卵障碍型不孕,而其促排卵作用可能部分是通过调节Smad4的表达,进而调控卵巢TGF-β/smads信号通路的激活来实现的。
OBJECTIVE: To observe the morphological changes of ovary and the significance of expression of Smad4 in TGF-β / smads pathway of Tongmaishengsheng Tablet. Methods Rat model of infertility with renal dysfunction of ovulation (ASR) was established. The rats were sacrificed after 5 weeks of successful modeling by vaginal smear. The rats were killed and the morphological changes were observed by HE staining. Western-blot ) To detect the expression of Smad4 protein in rat ovary. Results 1 The morphological changes of ovary in each treatment group were obvious. Mature follicles and corpus luteum were observed under microscope. Compared with the model group, the expression of Smad4 in the high, middle and low doses of Tongmaishengsheng tablet group and the positive control group increased (P <0.05), especially in the high-dose and letro-zole groups. Conclusion Tongmaishengsheng tablet can be used to treat infertility with renal dysfunction of ovulation, and its role in promoting ovulation may be partly through regulating the expression of Smad4, and then regulating the activation of ovarian TGF-β / smads signaling pathway to achieve.
