人类高分辨染色体(High-resoluti-on chromosome)是八十年代人类细胞遗传学技术发展的一个重要标志。这一技术的出现,不仅加强了人们对染色体细微结构的了解,而且有助于人们对由于染色体结构的细微改变所引起的疾病的认识。从1983年下半年开始,我们先后采用AO(Acridine orange,AO)法和AM-D(Actinomycin D,AMD)掺入法制备人类高分辩染色体,取得了较为满意的效果。现介绍如下: 材料和方法 (一)材料 1.RPMI 1640(日本产)2.小牛血请(上海产)3.PHA(自制)4.肝素5.Acrdine orange(Fluka) 6.Acti-nomycin D (Fluka)7.人体外周血。
High-resoluti-on chromosomes are an important symbol of the development of human cytogenetics in the 1980s. The advent of this technique not only reinforces people’s knowledge of the fine structure of chromosomes, but also helps people to understand the diseases caused by subtle changes in the structure of chromosomes. Since the second half of 1983, we have successfully prepared human high-resolution chromosomes using AO (Acridine orange, AO) and AM-D (AMD) incorporation methods, and achieved satisfactory results.
