目的 探索一种简单、敏感、特异和成本低的HIV-1亚型亚型测定方法。方法 采用改良的异源双链泳动法(HMA)与DNA序列分析法对14份HIV-1抗体阳性者样本进行亚型测定、分析和比较。结果 用HMA改良法明确测定了14份样本中的13份样本的亚型。在14份样本中对13份样本进行DNA序列分析有12份的结果与HMA的相符,符合率达92.3%。结论 改良的HMA测序法是一种简单、敏感、特异和经济的HIV-1亚型测定方法,可在广大的基层单位推广应用。
Objective To explore a simple, sensitive, specific and low cost HIV-1 subtype assay. Methods A total of 14 samples of HIV-1 positive were assayed, analyzed and compared by modified heteroduplex (HMA) and DNA sequencing. Results The subtypes of 13 of the 14 samples were clearly determined using the HMA modification method. In 14 samples of 13 samples of DNA sequence analysis, 12 of the results and the HMA consistent with a rate of 92.3%. Conclusion The improved HMA sequencing method is a simple, sensitive, specific and economical method for the determination of HIV-1 subtypes, which can be widely applied in grassroots units.
