南川百合(Lilium rosthornii Diels)是我国特有的野生百合种,可以作为抗逆育种的亲本,但是目前数量急剧减少。对重庆南川和湖南怀化的南川百合群体的生境及表型性状进行调查,测量形态学指标并计算其变异系数。从73对SSR引物中筛选出6对引物,对两个群体的遗传多样性进行分析。6对引物在两个群体中共扩增了15个条带,其中多态性条带为11条;根据Popgene计算可得,南川百合两个群体的遗传多样性在其群体平均水平为PPF=67.44%,N_e=1.684 5,I=0.544 9,H_(pop)=0.3507,物种水平为PPF=73.33%,N_e=1.730 2,I=0.612 1,H_(pop)=0.390 8;其中重庆群体的遗传多样性要高于湖南群体。两个群体之间的基因流为2.174 5,存在着基因交流;群体间遗传变异占10.31%,群体内的遗传变异占89.60%。作者对南川百合野生群体进行调研并对其群体遗传多样性进行初步研究,分析其濒危的原因,从而有利于南川百合保护、引种和育种方针的制定。
Lilium rosthornii Diels (Lilium rosthornii Diels) is a endemic wild lily of our country that can be used as the parent of anti-retrograde breeding, however, the number is rapidly decreasing. Habitats and phenotypic traits of Nanchuan lily population in Nanchuan, Chongqing and Huaihua, Hunan Province were investigated. Morphological indexes were measured and their coefficients of variation were calculated. Sixty-three primer pairs were screened from 73 pairs of SSR primers and their genetic diversity was analyzed. Sixteen pairs of primers amplified 15 bands in two populations, of which 11 were polymorphic bands. According to Popgene calculations, the genetic diversity of two populations of Nanchuan Lily was at the population level of PPF = 67.44 %, N_e = 1.684 5, I = 0.544 9, H_ (pop) = 0.3507, and the species level was PPF = 73.33%, N_e = 1.730 2, I = 0.612 1 and H_ (pop) Diversity is higher than the Hunan group. Gene flow between the two groups was 2.174 5, there was gene exchange; genetic variation among the groups accounted for 10.31%, and genetic variation within the population accounted for 89.60%. The author investigated the wild populations of Nanchuan Lily and conducted preliminary studies on the genetic diversity of their populations, and analyzed their reasons for endangerment so as to be beneficial to the protection, introduction and breeding of Nanchuan lilies.
