目的:探讨不同浓度的胰岛素和葡萄糖对TNF-α诱导单核细胞系U937可溶性ICAM-1表达的影响。方法:采用正交试验方法,将U937细胞置于含不同浓度(0、20、30、40和100mU/L)的胰岛素和不同浓度(5、10、20和30mmol/L)的葡萄糖及20μg/LTNF-α的RPMI1640培养液中培养。18h后,取培养液用ELISA法测定可溶性ICAM-1的水平。结果:U937细胞表达可溶性ICAM-1的水平随葡萄糖浓度的升高而增高,随胰岛素浓度的升高而下降。结论:胰岛素能抑制TNF-α诱导U937细胞表达可溶性ICAM-1。
Objective: To investigate the effects of different concentrations of insulin and glucose on TNF-α-induced monocyte cell line U937 soluble ICAM-1 expression. Methods: U937 cells were treated with different concentrations of insulin (0, 20, 30, 40 and 100 mU / L) and glucose (5, 10, 20 and 30 mmol / L) LTNF-α in RPMI1640 culture medium. After 18h, culture medium was used to determine the level of soluble ICAM-1 by ELISA. Results: The level of soluble ICAM-1 in U937 cells increased with the increase of glucose concentration and decreased with the increase of insulin concentration. Conclusion: Insulin can inhibit the expression of soluble ICAM-1 induced by TNF-α in U937 cells.
