目的:探讨正常子宫内膜和子宫内膜样腺癌组织中LRP16的表达及其与人子宫内膜腺癌的相关性。方法:应用RT-PCR技术研究10例正常子宫内膜组织及26例不同分化程度的子宫内膜样腺癌组织中LRP16 mRNA的表达;采用免疫组织化学方法检测上述子宫内膜癌组织中LRP16蛋白产物表达部位及表达水平。结果:子宫内膜样腺癌组织中LRP16mRNA的阳性表达率及表达水平(分别为83.33%、0.82±0.21)明显高于正常子宫内膜组织(分别为30%、0.47±0.18),P<0.05。LRP16在26例子宫内膜癌组织细胞核或胞浆中呈阳性表达,核阳性表达率为73.08%,浆阳性表达率96.15%。在不同分化程度的子宫内膜癌组织中:LRP16核表达与癌细胞的分化程度呈负相关(P<0.05),而LRP16浆表达与癌细胞的分化程度无相关性(P>0.05)。结论:子宫内膜癌组织中LRP16 mRNA的表达明显高于正常子宫内膜组织;且LRP16蛋白产物核表达水平与癌细胞的分化程度呈负相关;过高表达的LRP16与子宫内膜癌的发生和发展可能存在重要联系。
Objective: To investigate the expression of LRP16 in normal endometrium and endometrioid adenocarcinoma and its relationship with human endometrial adenocarcinoma. Methods: The expression of LRP16 mRNA in 10 cases of normal endometrium and 26 cases of endometrial adenocarcinoma with different degree of differentiation was studied by RT-PCR. The expression of LRP16 protein in endometrial carcinoma was detected by immunohistochemistry Product expression sites and expression levels. Results: The positive expression rate and expression level of LRP16 mRNA in endometrioid adenocarcinoma tissues were 83.33% and 0.82 ± 0.21, respectively, which were significantly higher than those in normal endometrial tissues (30% and 0.47 ± 0.18, P <0.05 respectively) . In endometrial carcinoma with different degrees of differentiation, the expression of LRP16 was negatively correlated with the differentiation of cancer cells (P <0.05), but there was no correlation between the expression of LRP16 and the differentiation of cancer cells (P> 0.05). CONCLUSIONS: The expression of LRP16 mRNA in endometrial carcinoma tissues is significantly higher than that in normal endometrial tissues.
