目的:观察比较关木通及当归四逆汤各拆方组中马兜铃酸A的变化趋势,探讨中药配伍对马兜铃酸A含量的影响。方法:参照当归四逆汤原方比例将供试样品分6组:关木通组、全方组、关木通+补气补血组、关木通+散寒组、关木通+甘草组、阴性对照组(全方去关木通);采用反相高效液相色谱(RP-HPLC)法,以甲醇—水—冰乙酸(体积比74∶24∶1)为流动相,以DiamonsilTM(钻石)C18(250 mm×4.6mm,5μm)为色谱柱,检测波长237 nm,测定各组中马兜铃酸A的含量。结果:与关木通组相比,当归四逆汤全方中马兜铃酸A含量最低;关木通+补血组马兜铃酸A含量也有明显降低。结论:在方剂配伍理论指导下,通过合理配伍降低关木通的毒性是可行的。
OBJECTIVE: To observe and compare the changes of aristolochic acid A in different prescriptions of Guan Mu Tong and Danggui Sini Decoction, and to explore the effect of traditional Chinese medicine compatibility on the content of aristolochic acid A. Methods: According to the proportion of the original part of Angelica Sini Decoction, the test samples were divided into 6 groups: Guanmutong group, Quanfang group, Guanmutong + Buqi Buxue group, Guanmutong + Sanxiong group, Guanmutong + licorice group and negative control group ; DiamonsilTM (diamond) C18 (250 mm × 4.6 mm, 5 μm) was prepared by reversed-phase high performance liquid chromatography (RP-HPLC) with methanol-water- For the column, detection wavelength of 237 nm, determination of aristolochic acid A content in each group. Results: Compared with Guanmutong group, the content of aristolochic acid A in Angelica Sini Decoction was the lowest, and the content of aristolochic acid A in Guanmutong + Xuexue group was also significantly decreased. Conclusion: Under the guidance of compatibility of prescriptions, it is feasible to reduce the toxicity of Guantong through rational compatibility.
