目的建立同时测定三黄颗粒中黄芩苷、小檗碱含量的HPLC法。方法采用Kromasil C18(4.6 mm×250 mm,5μm)色谱柱为固定相,以甲醇-0.1%磷酸水溶液为流动相,梯度洗脱,流速:1.0 mL.min-1,柱温:25℃,进样量:10μL,检测波长:黄芩苷为280 nm,盐酸小檗碱为345 nm。结果黄芩苷、小檗碱的线性范围分别为0.2048~2.0480μg(r=0.9999),0.2160~2.160μg(r=0.9999);平均加样回收率分别为99.89%、99.98%,RSD分别为1.45%、1.76%。结论本方法操作简便快捷,结果准确可靠,重复性好,可用于三黄颗粒的质量控制。
Objective To establish a HPLC method for the simultaneous determination of baicalin and berberine in Sanhuang Granules. Methods A Kromasil C18 column (4.6 mm × 250 mm, 5 μm) was used as the stationary phase. The mobile phase consisted of methanol-0.1% phosphoric acid solution with gradient elution at a flow rate of 1.0 mL · min- Sample volume: 10μL, detection wavelength: baicalin is 280 nm, berberine hydrochloride is 345 nm. Results The linear range of baicalin and berberine were 0.2048 ~ 2.0480μg (r = 0.9999) and 0.2160 ~ 2.160μg (r = 0.9999) respectively. The average recoveries were 99.89% and 99.98%, respectively, and the RSDs were 1.45% , 1.76%. Conclusion The method is simple, rapid, accurate, reliable and reproducible. It can be used for the quality control of Sanhuang granules.
