目的观察不同药物干预下,新生大鼠心肌细胞蛋白含量与活力的关系。方法采用考马斯亮蓝法检测蛋白含量,MTT比色法测量心肌细胞的A490值,以A490总细胞数之比作为衡量心肌细胞活力的指标。结果心肌细胞蛋白含量随着ET-1的浓度增大而增加,ET-1引起蛋白含量的增加可被拉西地平和川芎嗪减弱,心肌细胞的A490总细胞数之比随着ET-1的浓度增加而降低,拉西地平和川芎嗪也可进一步降低心肌细胞的A490总细胞数之比。结论心肌细胞蛋白质含量的增加并不引起细胞活力的增强,表明心肌细胞蛋白含量与活力之间可能缺乏相关关系。
Objective To observe the relationship between protein content and viability of neonatal rat cardiomyocytes under different drug interventions. Methods Protein content was determined by Coomassie brilliant blue method. The A490 value of myocardial cells was measured by MTT colorimetric method. The ratio of total A490 cells was used as a measure of myocardial viability. Results The protein content of cardiomyocytes increased with the increase of ET-1 concentration. The increase of protein content induced by ET-1 could be attenuated by lacidipine and tetramethylpyrazine. The ratio of the total number of A490 cells in cardiomyocytes with ET-1 Laxidipine and ligustrazine can further reduce the ratio of A490 total cell number of cardiomyocytes. Conclusion The increase of protein content in cardiomyocytes does not result in the increase of cell viability, indicating that there may be a lack of correlation between the protein content and viability of cardiomyocytes.
