目的:观察新加良附颗粒对人胃癌细胞SGC-7901移植瘤裸鼠血清IL-23、IL-21水平的影响,初步探讨其可能的作用机制。方法:60只裸鼠造模成功后随机分为空白组、5-Fu组、新加良附中剂量+5-FU组(联合给药组)及新加良附颗粒高、中、低剂量组。新加良附颗粒高、中、低剂量组给药剂量分别为10g·d~(-1)·kg~(-1)、5g·d~(-1)·kg~(-1)、2.5g·d~(-1)·kg~(-1);5-FU组给药剂量为17mg·d~(-1)·kg~(-1);空白组给予等体积生理盐水,联合给药组剂量为新加良附颗粒中剂量(5g·d~(-1)·kg~(-1))联合应用5-FU(17mg·d~(-1)·kg~(-1))。连续给药10天,进行眼球取血并处死小鼠,采用ELISA法检测血清IL-23、IL-21的含量。结果:联合给药组、5-FU组、新加良附高、中、低剂量组裸鼠血清中IL-23、IL-21含量较空白对照组均明显降低(P<0.01);与新加良附大剂量组、5-FU组比较,联合给药组血清IL-23含量差异无统计学意义(P>0.05),与新加良附大剂量组、5-FU组比较,联合给药组血清IL-21含量差异具有统计学意义(P<0.05)。结论:新加良附颗粒联合5-FU具有抑制人胃癌细胞SGC-7901移植瘤裸鼠血清中TH17细胞相关细胞因子IL-21、IL-23的表达,其可能通过调节TH17细胞相关细胞因子的表达影响胃癌的发生与发展。
OBJECTIVE: To observe the effects of Xinjialiangfu Granules on the levels of IL-23 and IL-21 in serum of human gastric cancer SGC-7901 transplanted tumor in nude mice, and to explore its possible mechanism. Methods: Sixty nude mice were randomly divided into blank group, 5-Fu group, medium dose plus 5-FU group (combination group) and Xinjialiang granules . The dosage of Xinjialiangfu granule was 10g · d -1 · kg -1, 5g · d -1 · kg -1, 2.5mg · d -1 · kg -1, g · d -1 · kg -1; the dosage of 5-FU group was 17 mg · d -1 · kg -1; the blank group was given equal volume of saline, The drug dose was 5g · d -1 · kg -1 in combination with 5-FU (17mg · d -1 · kg -1) . Continuous administration for 10 days, eyeball blood and mice were sacrificed, the contents of IL-23 and IL-21 in serum were detected by ELISA. Results: The levels of IL-23 and IL-21 in serum of nude mice in combination group, 5-FU group and Xingalaliang high, medium and low dose groups were significantly lower than those in blank control group (P <0.01) There was no significant difference in serum IL-23 levels between the two groups (P> 0.05). Compared with the high dose group of Galvanadine and the 5-FU group, the level of IL-23 in the combination group was not significantly different The difference of serum IL-21 level in the medicine group was statistically significant (P <0.05). Conclusions: Xinjiallian granule combined with 5-FU can inhibit the expression of TH17-related cytokines IL-21 and IL-23 in human gastric cancer SGC-7901 transplanted tumor nude mice by regulating the expression of TH17-related cytokines Expression affects the occurrence and development of gastric cancer.
