目的通过建立3T3中性红成纤维细胞光毒替代实验,研究体外替代方法替代动物实验及在化妆品安全性评价中取代动物光毒性实验的可行性。方法选用24种已知光毒性化学物质(15种阳性、9种阴性)和20种待检特殊用途化妆品,测量 Bal/c3T3细胞经化学物质和紫外线照射联合作用后的平均光效应强度和光刺激因子,与整体动物皮肤光毒性结果比较。结果 3T3体外替代实验对24种已知光毒性化学物质检测结果为14种阳性和10种阴性,20种化妆品为阴性,经校正 x~2检验与既往光毒性结果比较,差异无统计学意义。结论本次研究成功建立了3T3中性红成纤维细胞光毒替代实验方法,该方法可取代豚鼠皮肤光毒性实验对化妆品终产品进行安全性评价。
OBJECTIVE: To establish a 3T3 photophile alternative to neutral fibroblasts and to study the feasibility of replacing the animal experiment with an alternative method in vitro and to replace the phototoxicity experiment in cosmetics safety evaluation. Methods 24 kinds of known phototoxic chemicals (15 kinds of positive, 9 kinds of negative) and 20 kinds of special cosmetics to be tested were selected to measure the average light effect intensity and the light stimulating factor of Bal / c3T3 cells after the combination of chemical substances and ultraviolet radiation , Compared with the overall animal skin phototoxicity results. Results The results of 3T3 in vitro substitution tests showed that there were 14 positive and 10 negative results for 24 kinds of known phototoxic chemicals, and 20 kinds of cosmetics were negative. The corrected x ~ 2 test showed no significant difference with previous phototoxicity results. Conclusion This study successfully established a 3T3 neutral red fibroblasts photo-toxic alternative test method that can replace the guinea pig skin phototoxicity experiments for the safety evaluation of cosmetic final products.
