观察脂质体技术促进 c- myc反义寡核苷酸导入人体肝癌细胞的作用。方法 采用新一代脂质体 Lipofect AMINE( LR)包裹针对肝癌表达基因 c- myc反义寡核苷酸 ( ASODN) ,借助FITC荧光标记 c- myc- ASODN,应用细胞培养方法及荧光分光光度计以及荧光显微镜。结果 LR促进 ASODN进入靶细胞并增强其对肿瘤细胞基因的特异性封闭作用。结论 脂质体作为一种安全、简便、有效的基因转移载体广泛适用医学科研
The liposome technology was observed to promote the introduction of c-myc antisense oligonucleotides into human hepatoma cells. Methods A new generation of liposome Lipofect AMINE (LR) was used to coat the hepatoma-expressed c-myc antisense oligonucleotide (ASODN), with the help of FITC fluorescent labeling of c-myc-ASODN, using cell culture method and fluorescence spectrophotometer and Fluorescence microscope. Results LR promoted the entry of ASODN into target cells and enhanced its specific blocking effect on tumor cell genes. Conclusion Liposome is widely used in medical research as a safe, simple and effective gene transfer vector.
