深层培养古尼拟青霉小孢变种 (Paecilomycesgunnii var.minor Li etal.) PGM0 1菌株 ,在 5种不同的培养基中测试其菌丝体生物量。结果表明 ,以昆虫虫体为来源的蚕蛹粉培养基和松毛虫粉培养基较适合其生长 ,其中蚕蛹粉培养基为其深层发酵的最佳培养基。通过不同的方法分离纯化其多糖 ,表明 PGM0 1的胞内多糖有 3个组分 ,其主组分为淡褐色 ;具一般多糖的理化性质 ;气相色谱分析表明该多糖为甘露糖、葡萄糖和半乳糖组成的杂多糖 ,摩尔比约为 1 .5∶ 1 .3∶ 1。红外光谱分析表明 ,PGM0 1的胞内多糖主组分具有典型的多糖特征吸收峰
The Paecilomyces gunnii var. Minor Li et al. PGM0 1 strain was in-depth cultured and its mycelial biomass was tested in five different media. The results showed that the silkworm pupae culture medium and the pine caterpillar fungus medium suitable for insect growth were more suitable for their growth, of which silkworm pupa powder culture medium was the best medium for deep fermentation. The polysaccharides were separated and purified by different methods, which indicated that PGM0 1 had 3 components of intracellular polysaccharides and its main component was light brown. The physical and chemical properties of polysaccharides were general. The gas chromatographic analysis showed that the polysaccharides were mannose, glucose and half Lactose composition of heteropolysaccharide, the molar ratio of about 1.5: 1.3: 1. Infrared spectroscopy analysis showed that the main component of intracellular polysaccharide PGM0 1 has a typical polysaccharide characteristic absorption peak
