目的:观察补中益气丸对急性运动疲劳大鼠脑组织线粒体呼吸链复合体及ATP酶活性的影响。方法:将24只SD大鼠进行随机分组,分为安静对照组、急性运动组和补中益气丸组,每天按3g/kg的剂量灌胃补中益气丸混合液一次,药物补充一周后进行实验。急性运动为大强度间歇性跑台运动,速度为27m/min,坡度为0,总运动时间90min,每运动10min间歇1min。采用双抗体夹心法酶联免疫分析(ELISA)测定大鼠脑组织中线粒体呼吸链复合物活性浓度,采用无机磷法测定大鼠脑组织中Na+-K+-ATPase和Ca2+-ATPase活性浓度。结果:大鼠脑组织呼吸链复合体Ⅰ和Ⅲ各组之间均无显著性变化。与安静对照组比较,急性运动组呼吸链复合体Ⅱ活性、Na+-K+-ATPase和Ca2+-ATPase活性均显著降低;与急性运动组比较,补中益气丸组(3g/kg)呼吸链复合体Ⅱ、呼吸链复合体Ⅳ、Na+-K+-ATPase和Ca2+-ATPase活性均显著提高。结论:补中益气丸能改善急性运动后大鼠脑组织呼吸链复合体Ⅱ、Ⅳ及ATP酶活性。
Objective: To observe the effect of Buzhong Yiqi Pill on mitochondrial respiratory chain complex and ATPase activity in brain of acute exercise-induced fatigue rats. Methods: Twenty-four SD rats were randomly divided into three groups: control group, acute exercise group and Buzhongyiku pill group. The rats were treated with Buzhongyiqi mixture once daily at a dose of 3g / kg for one week After the experiment. Acute exercise for large-intensity intermittent treadmill speed, speed 27m / min, gradient 0, the total exercise time 90min, intermittent 1min every 10min exercise. The activity of mitochondrial respiratory chain complex in rat brain tissue was measured by enzyme-linked immunosorbent assay (ELISA). The concentrations of Na + -K + -ATPase and Ca2 + -ATPase in rat brain were measured by inorganic phosphorus method. Results: No significant changes were found in the respiratory chain complex Ⅰ and Ⅲ in rat brain tissue. Compared with the control group, the activities of respiratory chain complex Ⅱ, Na + -K + -ATPase and Ca2 + -ATPase in acute exercise group were significantly decreased. Compared with the acute exercise group, Buzhongyiqi pill group (3g / kg) Body Ⅱ, respiratory chain complex Ⅳ, Na + -K + -ATPase and Ca2 + -ATPase activity were significantly increased. Conclusion: Buzhong Yiqi Pill can improve the activities of respiratory chain complex Ⅱ, Ⅳ and ATPase in rat brain after acute exercise.
