目的:研究肾小管上皮细胞IL-4受体和IL-13受体的表达及IL-4和IL-13对肾小管上皮细胞形态和功能的影响,并探讨其作用机制。方法:借助人近端肾小管上皮细胞系(HK2),采用RT-PCR,免疫荧光方法检测HK2上IL-4受体和IL-13受体的表达。免疫荧光方法观察不同浓度IL-4和IL-13干预HK2细胞12~48h后HK2损伤标志物RANTES的变化情况。Western blot方法检测IL-4和IL-13干预HK2细胞后,JAK-STAT6信号通路的活化以及JAK-STAT6信号通路特异性抑制剂来氟米特对IL-4和IL-13作用HK2细胞的影响。结果:(1)RT-PCR和免疫荧光结果显示HK2有IL-4II型受体和IL-13I型受体mRNA和蛋白质表达。(2)IL-4和IL-13呈时间-剂量依赖性影响上皮细胞RANTES的表达。100ng/mlIL-4、50ng/mlIL-13作用48h对HK2的损伤作用最为明显。正常HK2细胞RANTES呈阴性,100ng/mlIL-4和50ng/mlIL-13干预48h后,阳性细胞显著增多。(3)Western blot方法结果显示JAK-STAT6磷酸化水平均在IL-4和IL-1350ng/ml作用10min时开始升高,IL-4100ng/ml作用20min达高峰,IL-1350ng/ml20min达高峰。JAK-STAT6信号通路特异性抑制剂来氟米特可阻断JAK-STAT6信号通路的活化,拮抗IL-4和IL-13诱导的HK2细胞RANTES表达。结论:(1)本研究首次证实人近端肾小管上皮细胞存在IL-4和IL-13受体的表达。(2)IL-4和IL-13可通过人近端肾小管上皮细胞上的相应受体造成细胞损伤,表现为RANTES表达增加。(3)IL-4和IL-13的上述作用与细胞内JAK-STAT6信号通路活化密切相关。
Objective: To investigate the expression of IL-4 receptor and IL-13 receptor in renal tubular epithelial cells and the effects of IL-4 and IL-13 on the morphology and function of renal tubular epithelial cells. Methods: The human proximal tubular epithelial cell line (HK2) was used to detect the expression of IL-4 receptor and IL-13 receptor on HK2 by RT-PCR and immunofluorescence. Immunofluorescence method was used to observe the changes of HK2 injury marker RANTES after intervention of different concentrations of IL-4 and IL-13 for 12-48 h in HK2 cells. The effect of JAK-STAT6 signaling pathway activation and leflunomide, a specific inhibitor of JAK-STAT6 signaling pathway, on HK2 cells induced by IL-4 and IL-13 were examined by Western blot after IL-4 and IL-13 intervention in HK2 cells . Results: (1) RT-PCR and immunofluorescence showed that HK2 had IL-4II receptor and IL-13I receptor mRNA and protein expression. (2) IL-4 and IL-13 affect the expression of RANTES in epithelial cells in a time-and dose-dependent manner. The effect of 100ng / ml IL-4 and 50ng / ml IL-13 on HK2 for 48h was the most obvious. Normal HK2 cells were negative for RANTES. After intervention with 100ng / ml IL-4 and 50ng / ml IL-13 for 48h, the number of positive cells increased significantly. (3) The result of Western blot showed that the level of JAK-STAT6 phosphorylation began to increase at 10min after IL-4 and IL-1350ng / ml, IL-4100ng / ml peaked at 20min, IL-1350ng / ml reached the peak at 20min. Leflunomide, a specific inhibitor of JAK-STAT6 signaling pathway, blocked the activation of JAK-STAT6 signaling pathway and antagonized IL-4 and IL-13-induced RANTES expression in HK2 cells. Conclusions: (1) This study for the first time confirmed the presence of IL-4 and IL-13 receptor expression in human proximal tubular epithelial cells. (2) IL-4 and IL-13 can cause cell injury through the corresponding receptors on human proximal tubular epithelial cells, showing an increase of RANTES expression. (3) The above effects of IL-4 and IL-13 are closely related to the activation of JAK-STAT6 signaling pathway in cells.
