目的探讨人重组干扰素-α2b(IFN-α2b)对尿道瘢痕成纤维细胞生长及其胶原合成的影响。方法建立尿道瘢痕成纤维细胞的体外培养模型,采用流式细胞术检测IFN-α2b(浓度分别为0、100、500、1 000 IU/mL,n=6)对其细胞周期的影响;采用羟脯氨酸比色法检测对其胶原合成量的影响。结果加入IFN-α2b 72 h后检测,随着加入IFN-α2b浓度的增加,①S-G2-M期细胞百分数降低(P<0.05);②尿道瘢痕成纤维细胞的胶原含量显著降低(P<0.05)。结论 IFN-α2b是成纤维细胞负性调节因子,降低胶原合成,可有效抑制尿道瘢痕形成。
Objective To investigate the effect of recombinant human interferon-α2b (IFN-α2b) on the growth of urethral scar fibroblasts and its collagen synthesis. Methods The in vitro culture model of urethral scar fibroblasts was established. The effects of IFN-α2b (0, 100, 500 and 1 000 IU / mL, n = 6) on the cell cycle were detected by flow cytometry. Proline Colorimetric Assay on Its Collagen Synthesis. Results The percentage of cells in S-G2-M phase decreased (P <0.05) with the increase of IFN-α2b concentration at 72 h after addition of IFN-α2b; ② The collagen content of scar fibroblasts in urethra decreased significantly (P <0.05 ). Conclusion IFN-α2b is a negative regulator of fibroblasts, reducing collagen synthesis, which can effectively inhibit urethral scarring.
