痛风性肾病 (goutynephropathy ,GN) ,即痛风肾 ,发病率呈上升趋势 ,是痛风致死的首要原因 ,目的 :探讨护肾痛风泰冲剂对模型大鼠痛风肾的效应机理。方法 :70只大鼠随机分为 4组 ,以腺嘌呤灌胃 ,复制痛风肾病理模型 ,给予相应药物。结果 :模型对照组肾重量、血清Ⅲ型前胶原 (PCⅢ )水平明显高于正常对照组 (P <0 .0 1)。护肾痛风泰组和别嘌呤醇组肾髓质和皮质内尿酸盐结晶比模型对照组明显减少 ,肾小管萎缩和肾小球减少程度均较模型对照组为轻。护肾痛风泰组血清PCIII水平较模型对照组明显降低 (P<0 .0 1) ,护肾痛风泰降低该模型大鼠的血清PCIII水平作用与别嘌呤醇相当 (P >0 .0 5 )。护肾痛风泰和别嘌呤醇对血清PCIII的降低水平接近正常对照组水平 (P >0 .0 5 )。结论 :复制大鼠痛风肾病理模型是成功的 ,护肾痛风泰能明显降低该模型大鼠血清PCIII水平。
Gouty nephropathy (GN), the gout nephropathy, has an increasing incidence and is the leading cause of gout death. Objective: To investigate the mechanism of the effect of Hushen Tongfengtai Granule on gout kidney in model rats. Methods: 70 rats were randomly divided into 4 groups. Adenine was intragastrically administered and the pathological model of gout kidney was duplicated. The corresponding drugs were given. Results: The kidney weight and serum PC III levels in the model control group were significantly higher than those in the normal control group (P < 0.01). The renal medulla and cortico-uric acid crystals in the kidney-protecting, goutytoin-group and allopurinol-treated groups were significantly lower than those in the control group, and tubular atrophy and glomerular reduction were milder than those in the model control group. Serum PCIII levels in Huishitongfengtai group were significantly lower than those in the model control group (P<0.01). Huoxuetongfengtai reduced the serum PCIII level of the model rats and was equivalent to allopurinol (P > 0.05). . The levels of serum PCIII in the patients with kidney and gout tetanus and allopurinol were similar to those in the normal control group (P > 0.05). Conclusion : The replication of rat gout kidney pathology model is successful, Hushen Tongfengtai can significantly reduce the level of serum PCIII in the rat model.
