目的:观察桑黄云芝胶囊对小鼠肉瘤S180及肝癌H22实体瘤的抑瘤作用及对肝癌H22腹水型小鼠生存期的影响。方法:昆明小鼠分别右腋皮下接种肉瘤S180及肝癌H22瘤种,连续灌胃给药12 d后处死动物,剥离皮下肿瘤并称重。S180瘤块用4%多聚甲醛固定,用免疫组织化学法检测瘤组织中血管内皮生长因子(VEGF),CD4及CD8的表达。另取昆明小鼠ip肝癌H22瘤种,连续桑黄云芝胶囊ip 10 d,停药后继续观察至小鼠死亡,记录小鼠生存天数,计算生命延长率。结果:与模型组相比,桑黄云芝胶囊可使S180和H22瘤重明显减轻,抑制VEGF表达,增强CD4和CD8表达,可明显延长H22肝癌腹水型荷瘤小鼠的生存天数。结论:桑黄云芝胶囊可抑制肿瘤生长,延长荷瘤小鼠生存期,其作用机理部分与抑制新生血管生成、增强机体免疫力有关。
OBJECTIVE: To observe the inhibitory effect of Sang Huangyunzhi capsule on the tumor-inhibiting effect of mouse sarcoma S180 and hepatocellular carcinoma H22 solid tumor and on the survival period of hepatoma H22 ascites type mice. METHODS: Kunming mice were inoculated subcutaneously with sarcoma S180 and hepatoma H22 tumors on the right iliac crest. Animals were sacrificed after continuous intragastric administration for 12 days. The subcutaneous tumors were excised and weighed. S180 tumor blocks were fixed with 4% paraformaldehyde. The expression of vascular endothelial growth factor (VEGF), CD4, and CD8 was detected by immunohistochemistry. Another Kunming mouse ip hepatocarcinoma H22 tumor type, continuous sanghuangyunzhi capsule ip 10 d, continue to observe until the mice died after drug withdrawal, record the number of days of survival mice, calculate the life extension rate. Results: Compared with the model group, Sang Huangyunzhi capsule can significantly reduce the weight of S180 and H22 tumors, inhibit the expression of VEGF, enhance the expression of CD4 and CD8, and significantly prolong the survival days of H22 ascites tumor-bearing mice. Conclusion: Sanghuang Yunzhi Capsule can inhibit tumor growth and prolong the survival period of tumor-bearing mice. Its mechanism of action is partly related to inhibition of neovascularization and enhancement of immunity.
