目的:建立测定蒙医烘制草乌中的乌头碱含量的高效液相色谱法。方法:采用高效液相色谱法测定蒙医烘制草乌中的乌头碱含量。色谱条件:色谱柱为Agilent EclipseXDB-C18(4.6 mm×250 mm,5μm);以乙腈-四氢呋喃(25∶15)为流动相A,以0.1 mol/L醋酸铵溶液(每1000 mL加冰醋酸0.5 mL)为流动相B(25∶75);流速为1.0 mL/min;检测波长为235 nm;柱温为室温。结果:乌头碱浓度在12.96~77.76μg/mL范围内与峰面积线性关系良好(r=0.9998),回归方程为Y=12466X+2383.9;加样回收率为103.09%,RSD为0.88%。理论塔板数按乌头碱峰计算不低于2000。结论:本方法具有简便、快速、准确的优点,可以作为蒙医烘制草乌乌头碱含量的测定方法。
Objective: To establish a high performance liquid chromatographic method for the determination of aconitine in Mongolian medicine. Methods: The content of aconitine in Mongolian medicine was determined by high performance liquid chromatography. Chromatographic conditions: Agilent EclipseXDB-C18 column (4.6 mm × 250 mm, 5 μm); acetonitrile-tetrahydrofuran (25:15) as mobile phase A, 0.1 mol / L ammonium acetate solution mL) as mobile phase B (25:75), flow rate 1.0 mL / min, detection wavelength at 235 nm and column temperature at room temperature. Results: The linear range of aconitine was 12.96 ~ 77.76μg / mL (r = 0.9998). The regression equation was Y = 12466X + 2383.9. The recoveries were 103.09% with RSD of 0.88%. The number of theoretical plates calculated aconitine peak of not less than 2000. Conclusion: The method has the advantages of simple, rapid and accurate, and can be used as a method for the determination of aconitine in baker’s yeast.
