本研究旨在探讨体外循环中红细胞损伤与氧自由基及钙的关系。资料与方法非紫绀型先天性心脏病患儿24例,分别于转流前、转流3分钟、转流结束、转流后2小时、24小时采集肝素抗凝血测定:①红细胞钙(E-Ca~(2+)):原子分光光谱法。②红细胞膜过氧化脂质(Em-LPO):硫代巴比妥酸法。③红细胞膜Ca~(2+),Mg~(2+)-ATP酶(EmCa~(2+),Mg~(2+)-ATPase)和红细胞膜Na~+,K~+-ATP酶(Em-Na~+,K~+-ATPase):同步测定法。④血浆游离血红蛋白(PF-Hb):邻联甲苯胺法。⑤应用电子显微镜观察红细胞形态,定量测定其面积(E-SA)和体积(E-Vol)。PF-Hb受转流中血液稀释的影响,用HCT校正:校正值=转流前HCT/实测时HCT×实测值
This study aimed to investigate the relationship between erythrocyte injury during cardiopulmonary bypass and oxygen free radicals and calcium. Materials and Methods 24 cases of non-cyanotic congenital heart disease in children, respectively, before the diversion, diversion for 3 minutes, the end of the commutation, 2 hours after the transfer of heparin anticoagulant 24 hours collected: ① erythrocyte calcium (E -Ca ~ (2+)): atomic spectrometry. ② erythrocyte membrane lipid peroxidation (Em-LPO): thiobarbituric acid method. ③ The changes of Ca ~ (2 +), Mg ~ (2 +) - ATPase (EmCa ~ (2 +) and Mg ~ (2 +) - ATPase) and erythrocyte membrane Na ~ + and K ~ + -ATPase Em-Na ~ +, K ~ + -ATPase): synchronous assay. ④ plasma free hemoglobin (PF-Hb): o-tolidine method. ⑤ The morphology of erythrocytes was observed with electron microscope, and the area (E-SA) and volume (E-Vol) were quantitatively determined. PF-Hb is affected by hemodilution in the bypass, corrected with HCT: Correction value = HCT before commutation / HCT measured during actual measurement
