目的探讨COX-2和MMP-9在人胃癌组织中的表达及临床意义。方法应用免疫组化SABC法检测45例人胃癌组织中COX-2、MMP-9的表达,并以30例正常胃黏膜组织作为对照。对COX-2、MMP-9的表达采用平均光密度测定,并对临床相关资料进行统计学分析。结果胃癌组织中COX-2、MMP-9的平均光密度值(OD)为19.85±7.34,18.81±7.56,显著高于正常胃黏膜组织的表达(12.49±4.87,11.46±4.82,P<0.01)。癌组织中COX-2、MMP-9的表达与患者性别、年龄及肿瘤的部位、组织类型、分化程度无相关性,而与肿瘤直径、TNM分期、淋巴转移、远处转移相关(P<0.05);同时经相关分析发现,在胃癌组织中,COX-2与MMP-9表达有显著相关性(r=0.437,P=0.001)。结论COX-2可能诱导MMP-9的表达而促进肿瘤的侵袭转移。
Objective To investigate the expression and clinical significance of COX-2 and MMP-9 in human gastric cancer. Methods The expression of COX-2 and MMP-9 in 45 cases of human gastric carcinoma was detected by immunohistochemical SABC method. Thirty cases of normal gastric mucosa were used as control. The average optical density of COX-2 and MMP-9 was measured, and the clinical data were analyzed statistically. Results The average OD values of COX-2 and MMP-9 in gastric cancer tissues were 19.85 ± 7.34 and 18.81 ± 7.56, respectively, which were significantly higher than those in normal gastric mucosa (12.49 ± 4.87, 1.146 ± 4.82, P <0.01) . There was no correlation between the expression of COX-2 and MMP-9 in cancer tissues, gender, age, tumor location, histological type and differentiation degree, but correlated with tumor diameter, TNM stage, lymph node metastasis and distant metastasis ). At the same time, the correlation analysis showed that there was a significant correlation between the expression of COX-2 and MMP-9 in gastric cancer (r = 0.437, P = 0.001). Conclusion COX-2 may induce the expression of MMP-9 and promote tumor invasion and metastasis.
