目的:探讨胚胎培养液中可溶性白细胞抗原-G(soluble leukocyte antigen-G,sHLA-G)的表达与早期胚胎发育的关系及其对IVF-ET妊娠结局的影响。方法:留取第3天移植及冷冻后的胚胎培养液,采用ELISA检测sHLA-G的表达水平。结果:1Ⅰ级胚胎与Ⅱ级胚胎培养液中sHLA-G表达水平相比较,差异无统计学意义(P>0.05),Ⅰ级和Ⅱ级胚胎培养液中sHLA-G表达水平均明显高于Ⅲ、Ⅳ级胚胎(P<0.05)。2双原核不同细胞数≤4细胞、5~6细胞、7~9细胞对应胚胎培养液中sHLA-G表达水平有增高趋势,但差异无统计学意义(P>0.05)。3妊娠组胚胎培养液中sHLA-G的表达水平显著高于非妊娠组(P<0.05)。结论:胚胎培养液中sHLA-G的表达水平可作为预测胚胎质量及妊娠结局的参考指标之一。
Objective: To investigate the relationship between soluble leukocyte antigen-G (sHLA-G) expression and embryonic development in embryo culture and its effect on IVF-ET pregnancy outcome. Methods: The embryonic culture fluid was collected from day 3 after transplantation and frozen. The expression of sHLA-G was detected by ELISA. Results: There was no significant difference in the level of sHLA-G expression between stage I and stage II embryos (P> 0.05). The levels of sHLA-G in stage I and stage II embryos were significantly higher than those of stage III , Grade IV embryos (P <0.05). The expression of sHLA-G in embryo culture fluid with different number of prokaryotic cells ≤ 4 cells, 5 ~ 6 cells and 7 ~ 9 cells had an increasing trend, but the difference was not statistically significant (P> 0.05). The expression level of sHLA-G in embryo culture group was significantly higher than that in non-pregnancy group (P <0.05). Conclusion: The expression level of sHLA-G in embryo culture fluid can be used as a reference index for predicting embryo quality and pregnancy outcome.
