直链N-磷酰氨基酸在水中会发生显著水解生成磷酸单酯、磷酸二酯、磷酸.其反应活性受酸度和侧链官能基的影响.在近中性条件下咪唑具有催化水解活性.环状N-磷酰氨基酸只生成磷酸双酯,中性条件下即使有咪唑催化也不反应.分子内羧基催化形成五元环状五配位中间体是反应的关键.
