目的:探讨黑素瘤抗原(melanoma antigen,MAGE)-A11在乳腺癌组织及相应癌旁组织中的表达,以及MAGE-A11对乳腺癌细胞增殖的影响。方法:应用免疫组织化学法检测60例乳腺癌组织及其相应癌旁组织中MAGE-A11蛋白的表达,分析其与乳腺癌患者临床病理学特征之间的关系。将pCMV-AC-MAGE-A11-GFP表达质粒转染人乳腺癌MCF-7细胞后,应用MTT法及集落形成实验检测MAGE-A11对乳腺癌细胞增殖的影响。结果:60例乳腺癌组织中有31例MAGE-A11蛋白阳性表达,阳性率为51.7%,相应的癌旁组织中均未发现MAGE-A11蛋白的表达;MAGE-A11蛋白的表达与乳腺癌患者的年龄、病理类型、组织学分级、临床分期、肿瘤大小、淋巴转移、雌激素受体(estrogen receptor,ER)-α、孕激素受体(progestrogen receptor,PR)及乳腺癌扩增因子1(ampli?ed in breast cancer1,AIB-1)的表达均无明显关系(P>0.05),与人表皮生长因子受体2(human epidermal growth factor receptor2,HER-2)/C-erbB2和ER-β的表达有关(P<0.05)。转染pCMV-AC-MAGE-A11-GFP表达质粒后,MCF-7细胞的增殖率和集落形成数均有明显增加(P<0.05)。结论:乳腺癌组织中MAGE-A11蛋白的表达与C-erbB2和ER-β的表达有关,且MAGE-A11可促进人乳腺癌MCF-7细胞的增殖。
Objective: To investigate the expression of melanoma antigen (MAGE) -A11 in breast cancer tissues and corresponding paracancerous tissues and the effect of MAGE-A11 on the proliferation of breast cancer cells. Methods: Immunohistochemistry was used to detect the expression of MAGE-A11 protein in 60 cases of breast cancer tissues and corresponding paracancerous tissues. The relationship between MAGE-A11 protein and the clinicopathological characteristics of breast cancer patients was analyzed. After transfection of pCMV-AC-MAGE-A11-GFP expression plasmid into human breast cancer MCF-7 cells, the effect of MAGE-A11 on the proliferation of breast cancer cells was detected by MTT assay and colony formation assay. Results: The positive expression rate of MAGE-A11 protein in 31 of 60 breast cancer tissues was 51.7%. The expression of MAGE-A11 protein was not found in the corresponding paracancerous tissues. The expression of MAGE- Age, pathological type, histological grade, clinical stage, tumor size, lymphatic metastasis, estrogen receptor (ER) -α, progestrogen receptor (PR) and breast cancer amplification factor 1 (P> 0.05), but not with the expression of HER-2 / C-erbB2 and ER-β (P <0.05). After transfection with pCMV-AC-MAGE-A11-GFP plasmid, the proliferation rate and colony formation of MCF-7 cells were significantly increased (P <0.05). Conclusion: The expression of MAGE-A11 in breast cancer tissues is related to the expression of C-erbB2 and ER-β, and MAGE-A11 can promote the proliferation of human breast cancer MCF-7 cells.
