目的:观察熟地(radix rehmanniae preparata)与泽泻(Rhizoma Alismatis)组成的药对在不同剂量配比下对庆大霉素(GM)损伤毛细胞的不同保护作用,寻找其最佳剂量配比。方法:复制内耳螺旋器体外培养GM损伤模型,参照基线等比增减设计,对熟地、泽泻设8个剂量配比组,通过耳蜗毛细胞计数观察不同剂量配比下熟地、泽泻对GM损伤耳蜗内、外毛细胞损失率的影响。结果:全耳蜗计数结果显示,熟地泽泻在8∶3、8∶8、0∶3、0∶8配比下,外毛细胞损失率在全耳蜗范围内较GM模型组有明显下降(P<0.01)。其他配比组与模型组无统计学差异。结论:熟地泽泻在不同配比下对毛细胞损失率的影响呈现出一种非线性的量效关系,原方配比(8∶3)及其他配比(8∶8、0∶3、0∶8)属最佳配比。
OBJECTIVE: To observe the different protective effects of radix rehmanniae preparata and Rhizoma Alismatis on hair cells damaged by gentamicin (GM) in different doses and to find the best dose ratio. Methods: The model of GM injury was established by replicating the inner ear spiral in vitro. According to the design of the baseline increase and decrease, eight doses of Radix Rehmanniae and Alisma were randomly divided into eight groups. Effect of injury on cochlear inner and outer hair cell loss rate. Results: Whole-cochlear counting results showed that the external hair loss rate of Alzheimer’s disease was significantly decreased in the whole cochlea than that in the GM model at the ratio of 8: 3, 8:8, 0:3 and 0:8 (P <0.01). Other matching group and the model group no significant difference. Conclusion: Rhizoma Alismatis and Radix Rehmanniae Preparata have a non-linear dose-and-effect relationship with different ratio of hair loss rate, the ratio of the original prescription (8: 3) and other ratio (8: 8, 0: 0: 8) is the best ratio.
