应用放射免疫分析法(RIA)研究了第二、三作者在丹麦制备的6种抗牛胰岛素原单克隆抗体(McAb)的某些特点。结果表明:①6种McAb的滴度及亲和力分别为0.5~6×10~5和2.6~8.5×10~8M~(-1),RIA的灵敏度在0.15~8.0ng之间;②其中5种可与牛胰岛素原C肽结合,但与牛胰岛素未见反应,提示其抗原决定簇均位于前者的部分;③另一种McAb(007)与两者均无反应,推测其抗原决定簇可能位于连接牛胰岛素和C肽的四个氨基酸的三维空间结构区.除007与猪胰岛素原有明显交叉反应外,其余的与人和猪胰岛素原均无交叉反应.上述抗体可用于牛胰岛素原的纯化、新的免疫鉴定法的建立及区分其种属来源等方面.
Radioimmunoassay (RIA) was used to study some of the six anti-bovine proinsulin monoclonal antibodies (McAbs) prepared by the second and third authors in Denmark.
