目的 探讨N ras反义寡脱氧核苷酸 (ASODN)对前列腺癌细胞增殖的影响。方法 针对N ras基因转录、翻译起始区分别合成硫代修饰的ASODN ,采用3H TdR掺入、流式细胞荧光免疫和斑点杂交法检测LNCaP细胞增殖和N ras基因表达。结果 两种ASODN单独作用后 ,LNCaP细胞内N rasmRNA、p2 1蛋白和PSA表达水平以及3H TdR掺入率明显低于对照组 ,10 μmol/L浓度对细胞生长的抑制率分别达 63 % ,5 1% ;二者联合应用后 ,N ras基因表达、PSA水平以及3H TdR掺入率进一步降低 ,细胞生长抑制率进一步增高达 85 %。结论 N rasASODN可有效抑制前列腺癌细胞增殖 ;同时证明N ras在前列腺癌发生发展中起重要作用
Objective To investigate the effect of NsRNA antisense oligodeoxynucleotide (ASODN) on the proliferation of prostate cancer cells. Methods Thio-modified ASODNs were synthesized in transcription and translation initiation regions of N ras respectively. 3H-TdR incorporation, flow cytometry immunofluorescence and dot blot hybridization were used to detect the proliferation and N ras gene expression in LNCaP cells. Results After treated with ASODN alone, the expression of N rasmRNA, p21 protein and PSA, and 3H TdR incorporation in LNCaP cells were significantly lower than those in control cells. The inhibition rates of 10 μmol / L and 10 μmol / L were 63%, 5% 1%. When combined, the N ras gene expression, PSA level and 3H TdR incorporation rate were further reduced, and the cell growth inhibition rate was further increased by 85%. Conclusion N ras ASODN can effectively inhibit the proliferation of prostate cancer cells; at the same time N ras also play an important role in the development of prostate cancer
