正常成人外周血细胞经二步法培养,可得到90%以上纯度的红系细胞。并且红系细胞随培养时间延长,不断向成熟分化,红系细胞数目增多。以逆转录多聚酶链反应法检测该培养体系内珠蛋白链mRNA相对含量。结果表明:随着红系细胞成熟分化,细胞内β-mRNA表达升高,而γ-mRNA表达下降。表明二步法建立的红系细胞液体培养较好地模拟了红系细胞发育的生理状态,为体外研究珠蛋白基因转换提供了良好模型。
Normal adult peripheral blood cells by two-step culture can be obtained more than 90% purity of erythroid cells. And erythroid cells with culture time, continue to differentiate into mature erythroid cells increased. Reverse transcriptase polymerase chain reaction was used to detect the relative content of mRNA of the inner globin chain in this culture system. The results showed that as erythroid cells mature and differentiated, the intracellular β-mRNA expression increased, while the γ-mRNA expression decreased. The results indicated that the liquid culture of erythroid cells established by the two-step method better simulated the physiological state of erythroid development and provided a good model for the study of globin gene transformation in vitro.
