目的:研究荷叶铁线蕨对人胚肝细胞增殖和功能的影响。方法:人胚肝细胞L-O2 96孔板104/孔培养,经质量浓度为1 g.mL-1荷叶铁线蕨水提液梯度稀释后处理24 h,采用MTT法检测细胞增殖情况;溴甲酚绿比色法检测白蛋白分泌水平;酶标仪法测定乳酸脱氢酶(LDH)活力。结果:荷叶铁线蕨水提液可以促进人胚肝细胞增殖,且具有剂量依赖性;在药物质量浓度为100 g.L-1时细胞白蛋白分泌量达到最高2.14 g.L-1,并显著降低了LDH的活力(P<0.05)。结论:荷叶铁线蕨水提液可以促进人胚肝细胞增殖,增强细胞保肝降酶作用。
Objective: To study the effects of Lonicera japonica on the proliferation and function of human embryonic hepatocytes. Methods: Human embryonic hepatocyte L-O2 96 / well 104 / well was cultured in a gradient concentration of 1 g.mL-1 for 24 h. The cell proliferation was detected by MTT assay. Bromocresol green colorimetric assay albumin secretion level; microplate reader determination of lactate dehydrogenase (LDH) activity. RESULTS: The water extract of Lonicera japonica Thunb. Could promote the proliferation of human embryonic hepatocytes in a dose-dependent manner. When the drug concentration was 100 gL-1, the secretion of albumin reached the highest level of 2.14 gL-1, and significantly decreased LDH activity (P <0.05). CONCLUSION: The water extract of Lonicera japonica Thunb. Can promote the proliferation of human embryonic hepatocytes and enhance the activity of liver cells to reduce the enzymes.
