目的分析健胃愈疡颗粒(JWYY)对乙酸性胃溃疡复发大鼠蛋白质表达谱的影响,寻找JWYY抗胃溃疡复发作用的相关蛋白。方法采用二维凝胶电泳(2-DE)分离JWYY处理组和JWYY未处理组大鼠胃组织的总蛋白,图像分析识别差异表达的蛋白质点,基质辅助激光解吸电离飞行时间质谱得到相应的肽质量指纹图,NCBInr及SWISS-PROT数据库搜索鉴定蛋白质。结果所获2-DE图谱分辨率高、重复性好,质谱分析共鉴定了12个差异蛋白质点,其中JWYY处理组有8个蛋白质点表达上调,4个蛋白质点表达下调,大多数蛋白与溃疡复发的形成、生长、凋亡及免疫等环节相关。结论JWYY抗胃溃疡复发作用可能与多种蛋白直接或间接相关,这些蛋白质的鉴定及进一步研究有助于抗溃疡复发药物新型靶标的发现。
Objective To analyze the effect of Jianweiyuyang granules (JWYY) on the protein expression profiles of recurrent rats with acetic acid-induced gastric ulcer, and to search for JWYY-related proteins that can prevent the recurrence of gastric ulcer. Methods Two-dimensional gel electrophoresis (2-DE) was used to separate the total protein of stomach tissue from JWYY treated and untreated JWYY rats. Image analysis was performed to identify differentially expressed protein spots. Matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) was used to obtain peptides. Mass fingerprinting, NCBInr and SWISS-PROT database search for identified proteins. Results The 2-DE map obtained had high resolution and good repeatability. Twelve differential protein spots were identified by mass spectrometry. Among them, 8 protein spots were up-regulated and 4 protein spots were down-regulated in JWYY treatment group. Most proteins and ulcers The formation, growth, apoptosis, and immunity of relapse are related. Conclusion The recurrence of JWYY against gastric ulcer may be directly or indirectly related to many proteins. The identification and further research of these proteins are helpful for the discovery of novel anti-ulcer drugs.
