目的考察通络醒脑泡腾片对东莨菪碱致拟痴呆小鼠学习记忆及中枢胆碱能神经功能的影响。方法制备东莨菪碱致拟痴呆小鼠模型,采用避暗法和Morris水迷宫考察通络醒脑泡腾片对小鼠模型学习记忆功能的影响;采用ELISA法检测小鼠脑海马组织乙酰胆碱(Ach)和胆碱乙酰转移酶(ChAT)含量,采用比色法测定AchE含量,考察通络醒脑泡腾片对小鼠模型中枢胆碱能神经功能的影响。结果避暗测定结果显示通络醒脑泡腾片可以显著延长小鼠的逃避潜伏期,并显著减少错误次数,与模型组比较差异有显著性意义(P<0.05);Morris水迷宫检测结果显示通络醒脑泡腾片能够显著缩短小鼠定向航行测试的逃避潜伏期和空间探索测试的第三象限停留时间,显著延长有效区停留时间,增加经过平台次数,与模型组比较差异有统计学意义(P<0.05);小鼠脑组织中枢胆碱能神经功能测定结果显示,模型组Ach含量显著降低,ChAT酶活性显著降低,AchE活性显著增高,与空白对照组比较差异有统计学意义(P<0.05);与模型组比较,通络醒脑泡腾片高、中、低剂量组Ach活性差异以及中剂量组ChAT活性显著增高,差异有统计学意义(P<0.05),而通络醒脑泡腾片AchE活性有一定的降低趋势,但差异无统计学意义(P>0.05)。结论通络醒脑泡腾片对东莨菪碱所致小鼠拟痴呆模型学习记忆功能具有良好的改善作用,可能与提高模型小鼠中枢胆碱能神经功能、促进乙酰胆碱的合成有关。
Objective To investigate the effect of Tongluo Xingnao effervescent tablet on learning and memory and central cholinergic function induced by scopolamine in mice. Methods Scopolamine-induced dementia mouse model was established. The effects of Tongluo Xingnao effervescent tablet on the learning and memory function of the mouse model were investigated by dark-avoidance method and Morris water maze. The levels of acetylcholine (Ach) and Choline acetyltransferase (ChAT) content, the determination of AchE content by colorimetric assay Tongluo Xingnao effervescent tablets in mouse model of central cholinergic nerve function. Results The results of dark avoidance test showed that Tongluo Xingnao effervescent tablet can significantly prolong the escape latency of mice and significantly reduce the number of errors, which is significantly different from the model group (P <0.05). Morris water maze test showed that Xingnao brain effervescent tablet can significantly shorten the escape latency and the third quadrant of the space exploration test, significantly prolong the residence time of the effective area and increase the number of passes through the platform, the difference was statistically significant compared with the model group ( P <0.05). The results of central cholinergic nerve function test showed that the content of Ach, the activity of ChAT and the activity of AchE in model group were significantly lower than those in control group (P < 0.05). Compared with the model group, the Ach activity of Tongluo Xingnao effervescent tablet high, medium and low dose groups and ChAT activity of middle dose group were significantly increased, the difference was statistically significant (P <0.05), while Tongluo Xingnao AchE activity of effervescent tablets has a certain downward trend, but the difference was not statistically significant (P> 0.05). Conclusions Tongluo Xingnao effervescent tablet can improve the learning and memory function of scopolamine-induced dementia model in mice, which may be related to improving central cholinergic function and promoting the synthesis of acetylcholine in model mice.
