目的在体外实验中下调gelsolin(GSN)的表达,观察其对孕晚期胎鼠心肌细胞增殖能力的影响。方法分离、原代培养孕15天SD胎鼠心肌细胞,采用干扰小RNA(small interference RNA,si RNA)方法,下调GSN在孕15天SD胎鼠心肌细胞中的表达;通过CCK-8实验检测胎鼠心肌细胞的增殖情况;采用流式细胞仪检测细胞周期分布的变化。结果 si RNA下调GSN的表达后,胎鼠心肌细胞的增殖能力下降(P<0.05);G0-G1期细胞比例高于对照组(P<0.05);S期细胞比例降低(P<0.05);差异有统计学意义。结论降低GSN基因的表达可以抑制细胞从G0-G1期进入S期,从而抑制胎鼠心肌细胞的增殖。
Objective To investigate the effect of gelsolin (GSN) on the proliferation of fetal rat cardiomyocytes in late pregnancy. Methods Primary cultured SD rat cardiac myocytes were isolated and cultured for 15 days. The expression of GSN in fetal SD rat cardiac myocytes was reduced by a small interfering RNA (siRNA) method. CCK-8 assay Fetal rat cardiomyocytes proliferation; using flow cytometry to detect changes in cell cycle distribution. Results The proliferation of fetal rat cardiomyocytes decreased after siRNA down-regulated the expression of GSN (P <0.05). The proportion of cells in G0-G1 phase was higher than that in control group (P <0.05). The difference was statistically significant. Conclusion Decreasing the expression of GSN gene can inhibit the cells from entering the S phase from the G0-G1 phase, thus inhibiting the proliferation of fetal rat cardiomyocytes.
