以白术提取物的得率及抗氧化活性为指标,选择甲醇体积分数、料液比、提取时间、提取温度为自变量,采用单因素实验和响应面法对白术提取工艺进行优化,结果表明:甲醇体积分数为87%、液料比35(mL/g)、提取温度为62℃、提取时间为1.5 h,在上述条件下白术得率为42.2322%,与模型的预期值42.2566%基本相符。对最佳的提取条件下的白术提取物进行抗氧化的测定,得出清除DPPH自由基的Ic50值为0.238 g/L,清除率比较好。
Atractylodes macrocephalae extraction technology was optimized by single factor experiment and response surface methodology with the yield and antioxidant activity of Atractylodes macrocephala extract as indexes. The optimal conditions were as follows: volume fraction of methanol, ratio of solid to liquid, extraction time and extraction temperature. The volume fraction of methanol was 87%, the ratio of liquid to solid was 35 (mL / g), the extraction temperature was 62 ℃, and the extraction time was 1.5 h. The yield of Atractylodes macrocephala was 42.2322% under the above conditions, which was basically consistent with the expected value of 42.2566%. Antioxidant activity of Atractylodes macrocephala extract under optimal extraction conditions was determined. The results showed that the Ic50 value of DPPH radical scavenging was 0.238 g / L and the clearance rate was good.
