本文以核孔膜过滤法纯化刚地弓形虫接种小鼠腹腔后所获腹水及其洗涤液中的速殖子。共纯化6批,约70ml含虫悬液。原虫回收率为65.8%,原虫纯度达98.87%。对宿主白细胞的清除率为98.4%,红细胞85.2%。宿主细胞占纯化后的原虫悬被中有形成份的比例均低于1%。过滤操作对速殖子的生命力、毒力和感染力等无影响。且一张滤膜可滤获10~8原虫。核孔膜具有孔形稳定、孔径均匀、分辨力高、低吸附和表面阻留等优点。该法是纯化弓形虫速殖子的稳定高效、省时简便的无损新方法。
In this paper, purified by nuclear membrane filtration Toxoplasma gondii inoculated mice peritoneal ascites and washing liquid tachyzoites. A total of 6 batches of purification, about 70ml containing insect suspension. Protozoa recovery was 65.8%, protozoa purity of 98.87%. The clearance rate of host white blood cells was 98.4% and erythrocytes 85.2%. Host cells account for less than 1% of the tangible components in the purified protozoan canopy. Filtering has no effect on tachyzoites viability, virulence and infectivity. And a filter can be filtered by 10 to 8 protozoa. Nuclear pore membrane with pore-shaped stability, uniform aperture, high resolution, low adsorption and surface retention and so on. This method is a stable and efficient purification of Toxoplasma gondii, saving time and simple new method of nondestructive.
