目的:比较盐酸阿霉素脂质体与盐酸阿霉素在大鼠体内组织中的分布特征。方法:大鼠股静脉注射盐酸阿霉素及盐酸阿霉素脂质体注射液(0.8mg·100g-1体质量),采用HPLC法测定不同时间点大鼠组织脏器中的药物浓度。结果:与盐酸阿霉素相比,盐酸阿霉素脂质体在心脏组织中具有较低的浓度,而在肝脏及肾脏组织中的药物浓度高于阿霉素。结论:将阿霉素制成以脂质体载药,可在一定程度上降低心脏毒副作用,提高药物对肝脏和肾脏等肿瘤的治疗效果。
OBJECTIVE: To compare the distribution characteristics of doxorubicin hydrochloride liposomes and doxorubicin hydrochloride in vivo in rats. METHODS: Rats were injected intravenously with doxorubicin hydrochloride and doxorubicin hydrochloride injection (0.8 mg · 100 g-1 body weight). The drug concentrations in the tissues of rats were measured by HPLC at different time points. RESULTS: Compared with doxorubicin hydrochloride, doxorubicin hydrochloride liposomes had a lower concentration in heart tissue and a higher concentration of drug in liver and kidney tissues than doxorubicin. CONCLUSION: Adriamycin can be delivered in liposomes to a certain extent, reduce the side effects of heart and improve the therapeutic effect of drugs on tumors such as liver and kidney.
