目的:探讨喉鳞状细胞癌组织中细胞外信号调节激酶(ERK)与尿激酶型纤溶酶原激活剂(uPA)表达间的关系,及其在喉鳞状细胞癌发展中的意义。方法:Western blot法检测40例喉鳞状细胞癌中ERK1/2蛋白的表达,同时检测ERK1/2酶活性以测定其磷酸转移酶效率。免疫组织化学方法检测相应组织中uPA蛋白表达。结合临床相关因素进行统计分析。结果:ERK1/2蛋白表达水平呈线性正相关(P<0.05),与T分级、病理学分级和颈淋巴结状况无关。ERK1/2酶活性表达水平和uPA蛋白表达水平与T分级及病理学分级无关;与颈淋巴结状况有关(P<0.05)。ERK1/2蛋白表达与ERK1/2酶活性表达及uPA蛋白表达无相关性;ERK1/2酶活性表达与uPA蛋白表达呈线性正相关(P<0.05)。结论:ERK激酶过度激活可能通过调控uPA表达上调,促进喉鳞状细胞癌的颈淋巴结转移。
Objective: To investigate the relationship between the expression of extracellular signal-regulated kinase (ERK) and urokinase-type plasminogen activator (uPA) in laryngeal squamous cell carcinoma and its significance in the development of laryngeal squamous cell carcinoma. Methods: Western blot was used to detect the expression of ERK1 / 2 protein in 40 laryngeal squamous cell carcinomas, and the activity of ERK1 / 2 was measured to determine its phosphotransferase efficiency. Immunohistochemistry was used to detect the expression of uPA protein in corresponding tissues. Combined with clinical factors related to statistical analysis. Results: The expression of ERK1 / 2 was positively correlated with the level of ERK1 / 2 (P <0.05), but not with the grade of T, pathological grade and the status of cervical lymph node. The expression level of ERK1 / 2 and uPA protein were not related with T grade and pathological grade, but with lymph node status (P <0.05). There was no correlation between the expression of ERK1 / 2 and the expression of ERK1 / 2 and uPA protein. The expression of ERK1 / 2 was positively correlated with the expression of uPA (P <0.05). Conclusion: Overexpression of ERK may up-regulate uPA expression and promote cervical lymph node metastasis in laryngeal squamous cell carcinoma.
