目的:研究补肾益气方及其拆方(黄芪、首乌)对氧化低密度脂蛋白(ox-LDL)所致动脉粥样硬化(AS)症血管平滑肌(VSMC)病理模型细胞的增殖及其调控基因的影响。方法:用ox-LDL处理VSMC复制“AS”细胞病理模型,用MTT法、流式细胞仪、RT-PCR法观察含受试药物血清对模型细胞增殖、泡沫细胞形成、增殖周期及相关基因表达影响。结果:补肾益气方及其拆方(黄芪、首乌)能降低模型细胞的增殖率及泡沫细胞形成率;降低S期细胞的百分率,下调VSMC增殖促进基因PDGF-a、c-MYC,上调增殖抑制基因P27的表达。结论:补肾益气方及其拆方具有抑制AS症的血管平滑肌细胞增殖和泡沫细胞的形成。
Objective: To investigate the effects of Radix Astragali, Radix et Rhizoma Rhei and Radix Astragali on the proliferation of vascular smooth muscle cells (VSMCs) induced by ox-LDL and its effects Regulate the impact of genes. Methods: VSMCs were stained with ox-LDL for “AS” cytopathological model. MTT assay, flow cytometry and RT-PCR were used to observe the effect of test drug serum on cell proliferation, foam cell formation, Effect of gene expression. Results: Bushen Yiqi recipe and its disassembled prescriptions (Astragalus and Radix Polygoni multiflori) could reduce the proliferation rate and foam cell formation rate of model cells, decrease the percentage of S phase cells and down-regulate the proliferation of PDGF-a and c-MYC Proliferation Inhibition Gene P27 Expression. Conclusion: Bushen Yiqi Fang and its disassembled prescriptions can inhibit the proliferation of vascular smooth muscle cells and the formation of foam cells in AS.
