目的优选左金丸最佳提取方法。方法采用超高效液相色谱法(UPLC),建立左金丸的指纹图谱以及5种生物碱的测定方法;以指纹图谱相似度和5种生物碱的总量作为评价指标,对左金丸的水提、70%乙醇提取、盐酸-甲醇(1∶100)提取、半仿生提取方法进行比较。结果左金丸的色谱峰在8 min内检测完,共检测到9个共有峰,基线平稳,峰形较好。5种生物碱的线性关系良好,分离度高;不同提取方法的左金丸指纹图谱相似度>0.98;而5种生物碱的量差异较大,其中盐酸-甲醇(1∶100)提取的左金丸中5种生物碱的总量最高,水提方法最低。结论本实验建立的方法操作简便,分析速度快;基于指纹图谱的定性判别和指标成分的定量分析,可以综合全面地评价比较左金丸的提取方法;其中盐酸-甲醇(1∶100)提取左金丸的方法优于其他3种方法。
Objective Optimization Zuo Jin Wan best extraction method. Methods The fingerprint of Zuojinwan and the determination of five alkaloids were established by ultra performance liquid chromatography (UPLC). The fingerprinting similarity and total amount of five alkaloids were used as indexes to evaluate the water extract of Zuojinwan , 70% ethanol extraction, hydrochloric acid - methanol (1: 100) extraction, semi-bionic extraction methods were compared. Results The chromatographic peaks of Zuojin pills were detected within 8 min. A total of 9 common peaks were detected. The baseline was stable and the peak shape was good. The five alkaloids had good linearity and high resolution. The similarity of Zuojin pill fingerprinting was> 0.98 with different extraction methods, while the amount of the five alkaloids was quite different. Among them, Zuojin pills extracted with hydrochloric acid-methanol (1: 100) The total amount of five kinds of alkaloids in the highest water extraction method is the lowest. Conclusion The method established in this experiment is simple and fast in analysis. Based on the qualitative identification of fingerprints and the quantitative analysis of index components, the extraction method of Zuojin pills can be comprehensively and comprehensively evaluated.
