目的:建立HPLC同时测定老鹳草属5种老鹳草中原儿茶酸、没食子酸、柯里拉京、鞣花酸和金丝桃苷含量的方法。方法:采用Agilent TC-C18色谱柱(4.6 mm×250 mm,5μm),流动相0.1%磷酸-乙腈梯度洗脱,流速1.0 m L·min-1,检测波长280 nm,柱温30℃。结果:原儿茶酸、没食子酸、柯里拉京、鞣花酸和金丝桃苷5种成分的线性范围分别为0.500~5.000μg(r=0.999 5),0.075~0.750μg(r=0.999 7),1.790~17.900μg(r=0.999 9),0.580~5.800μg(r=0.999 6),0.065~0.650μg(r=0.999 6),平均回收率在94.05%~102.58%,RSD在1.6%~2.9%。结论:该方法简单、准确,具有良好的重复性和稳定性,可为老鹳草药材的质量评价提供一定的参考依据。
Objective: To establish a HPLC method for the simultaneous determination of protocatechuic acid, gallic acid, corilagin, ellagic acid and hyperoside in five species of Geranium. METHODS: The mobile phase was eluted with 0.1% phosphoric acid-acetonitrile gradient on a TC-C18 column (4.6 mm × 250 mm, 5 μm) with a flow rate of 1.0 m L · min-1. The detection wavelength was 280 nm and the column temperature was 30 ℃. Results: The linear ranges of protocatechuic acid, gallic acid, corilagin, ellagic acid and hyperoside were 0.500 ~ 5.000μg (r = 0.999 5), 0.075 ~ 0.750μg (r = 0.999 7 ), 1.790 ~ 17.900μg (r = 0.999 9), 0.580 ~ 5.800μg (r = 0.999 6) and 0.065 ~ 0.650μg (r = 0.999 6) .The average recoveries ranged from 94.05% to 102.58% 2.9%. Conclusion: The method is simple and accurate, with good repeatability and stability, which can provide some references for the quality evaluation of geranium medicinal herbs.
